Cell Sorting in Earth's Microbial Frontiers

Happy cells = good science! High-pressure conditions of traditional FACS are known to damage cells, leading to cellular stress and decreased viability. NanoCellect’s low-pressure microfluidics systems purify target populations based on size and fluorescence parameters while maintaining high viability for wherever you send them next.

The WOLF G2 sorts undifferentiated induced pluripotent stem cells that grow into healthy colonies.

Immunocytochemistry of iPSCs on Emerging Colonies 6 Days Post-Sort

A. SSEA-4 AF488 (green) is a surface marker for pluripotency. B. Oct4 AF594 (red) is an intracellular marker to confirm pluripotency. C. DAPI (blue) is a marker used to confirm the number of nuclei of a cell. D. Overlay of a colony positive for SSEA-4, Oct4, and DAPI (10X, scale bar 170 µm).

The WOLF G2 Cell Sorter increases clonal outgrowth by up to 6 times compared to limiting dilution.

Average Single Cell Deposition When Using Limiting Dilution VS. WOLF G2

On average, the WOLF G2 with the N1 Single-Cell Dispenser resulted in a 4-6 fold increase of single cells that can potentially grow into colonies compared to limiting dilution at 1 cell/well under the same conditions.

In a new study, researchers aimed to elucidate the mechanism that prevents natural killer (NK) cells from effectively infiltrating solid tumors. By investigating the role of PDZ-binding kinase (PBK) in immune regulation, they discovered that inhibiting PBK can effectively increase the anti-tumor activity of NK cells. The WOLF cell sorter was used to isolate viable NK-92MI cells that were injected into tumors for downstream assays.
