Achieve higher clone quality and quantity by using single cell sorting.

We highlight the advantages that cell sorting can provide for developing monoclonal cell lines over the traditional technique, limiting dilution. We will review:

  •  What is limiting dilution, and what are its respective challenges and limitations
  • How cell sorting can enhance cloning efficiency, while reducing time, costs, and resources
  • How to increase the quality of clonal outcome by using fluorescent cell sorting
We showcase the quantitative differences between limiting dilution and cell sorting with respect to both seeding efficiency and clonal outgrowth. By utilizing the WOLF® Cell Sorter N1 Single Cell Dispenser to simultaneously sort and dispense single cells into 96- and/or 384-well plates, we can increase the overall number of clones/plate compared to limiting dilution.
